Well, let's not kid ourselves here - in principle this Gel Extraction/PCR Clean-up Kit behaves like others on the market and was designed to recover or concentrate DNA fragments (100 bp→10 Kb) from agarose gel, PCR, or other enzymatic reactions. What is great about these kits - no column fibres coming through to inhibit subsequent reactions or interfere with spec readings.
Chaotropic salt is used to dissolve agarose gel and inactivate enzymes. DNA fragments in the chaotropic salt are bound by the glass fibre matrix of the spin column. Contaminants are removed with a Wash Buffer (containing ethanol) and the purified DNA fragments are eluted by a low salt Elution Buffer. Salts, primer-dimers, enzymes and left-over dNTPs can be rapidly and effectively removed and low concentration DNA solutions effectively concentrated to smaller volumes.
